system, leading to a neosynthesis of ecdysone. A dose of 500 ng/pupa was sufficient to obtain synchronous adult development. Ecdysone injected either in whole or in decapitated pupae induced a neosynthesis of ecdysone, starting on day 5 after treatment, and the onset of imaginal development. Thus, we suggest that ecdysone can act independently of the brain, and that its stimulatory action on ecdysone neosynthesis is probably indirect, according to the long delay (5 days) necessary for this reactivation. In addition, isolated abdomens from diapausing pupae injected with the same dose of ecdysone also developed, but did not synthesize any ecdysteroid. This suggests that the injected hormone has also a direct morphogenetic effect on target tissues. The significance of this ecdysone neosynthesis is discussed.

Cellular changes in the prothoracic glands of diapausing pupae of Manduca sexta.

Prothoracic glands from diapausing pupae of the tobacco hornworm, Manduca sexta, synthesize markedly less ecdysone in vitro in response to prothoracicotropic hormone (PTTH) than do glands from non-diapausing pupae. Impaired steroidogenesis is also observed in glands from diapausing animals exposed to agents that enhance ecdysone synthesis in non-diapausing pupal glands by increasing intracellul...

Energy and lipid metabolism during direct and diapause development in a pierid butterfly.

Diapause is a fundamental component of the life cycle in the majority of insects living in environments characterized by strong seasonality. The present study addresses poorly understood associations and trade-offs between endogenous diapause duration, thermal sensitivity of development, energetic cost of development and cold tolerance. Diapause intensity, metabolic rate trajectories and lipid ...

Larval diapause termination in the bamboo borer, Omphisa fuscidentalis

In insects, juvenile hormone (JH) and 20-hydroxyecdysone (20E) regulate larval growth and molting. However, little is known about how this cooperative control is terminating larval diapause especially in the bamboo borer, Omphisa fuscidentalis. In both in vivo and in vitro experiments, we here measured the expression levels of genes which were affected by juvenile hormone analogue (JHA: S-metho...

Oxygen convective uptakes in gas exchange cycles in early diapause pupae of Pieris brassicae.

Oxygen convective uptakes in gas exchange cycles were directly recorded in early diapause pupae of Pieris brassicae L. (Lepidoptera; Pieridae) by means of O2 coulometric respirometry. This method was combined with flow-through CO2 respirometry, the two systems being switchable one to the other. During recording with both systems, measurements were also taken with infrared actography. The pupae ...

In Vitro Studies on the Role of the Brain and Prothoracic Glands in the Pupal Diapause of Manduca Sexta

Pupal diapause in the tobacco horn worm, Manduca sexta (Johannson), is characterized by the absence of an increased ecdysteroid titre in the haemolymph during the first week of pupal life as measured by radioimmunoassay. This virtual absence of the steroid moulting hormone is thought to be responsible for the diapause state and it is apparently a consequence of the failure of the diapausing pup...
